TMB (3,3´, 5,5´-tetramethylbenzidine)는 Peroxidase 촉매 작용에 의해 수소 과산화물로 산화될 때 파란색을 띄고 370 nm 및 652 nm에서 최대 OD값을 가지며, 황산등의 산에의해 반응을 정지시키면 노란색으로 바뀌며 450 nm에서 최대 OD값을 나타낸다.
Peroxichrom Excel^TM은 HRP(horseradish-peroxidase)가 부착된 Conjugate를 검출하기 위한 1- component TMB substrate로 기존에 사용하던 2-component 또는 Peroxichrom 과 비교했을 때 더욱 민감한 반응을 보인다. Peroxichrom Excel^TM substrate는 파란색일 때 650nm에서 최대 흡광도를 보이며, 반응 정지액(2N H₂SO₄)을 첨가한 후 노란색을 띄며 이때 450nm에서 최대 흡광도를 보인다.
Peroxichrom Excel^TM은 1-component로서 사용시 다른 부가적인 준비 없이 바로 사용할 수 있는 이점이 있으며 또한 높은 민감도를 보여 적은 양의 단백질이나 항체를 검출할 수 있다.
